Superdrol has one of the most fascinating stories in bodybuilding culture.

If I told many of you today that Superdrol was once sold on supplement shelves like a whey protein jar, it would probably sound like some kind of shenanigan — but yes, it was!

Not sure how many of you here are aware of it, or still remember it, but just like these days you hear about a new peptide that is supposed to revolutionize the fitness and health industry, back in the early 2000s there was the famous prohormone era, which started with compounds such as DHEA, a name many of you probably heard of.

Most of those so-called prohormones were quite a nothing in a capsule, or at least far less impressive than the hype around them.

Well, Superdrol was not one of them.

Superdrol is arguably one of the most potent steroids ever created, milligram per milligram. To make it even more appealing, it provides a rock-hard physique, and it is one of the most effective compounds for recomposition. It also provides impressive strength gains — which are especially remarkable considering the relatively mild mass gains it usually puts on one’s frame.

Of course, things come at a cost, and Superdrol is also quite liver toxic. But no worries, we’ll explore all of that below.

As always, we’ll start from its chemistry, and from there it will be much easier to build the understanding of how it acts and how it serves in sports and bodybuilding.

Superdrol, Methasterone, and the Masteron Connection

Superdrol is known by its generic name methasterone, but on PubMed you will find it under the chemical definition of methyldrostanolone, and this is quite telling.

Masteron is legendary in contest preparation, and it is one of the few drugs that may be used until the last day on stage, providing a sculpted, dense, vein-popping physique — exactly the one that pro bodybuilders are looking for.

Drostanolone / Masteron is a direct derivative of dihydrotestosterone, also known as DHT, the strongest natural androgen in the body. But unlike DHT, which has almost zero anabolic impact in muscle cells, drostanolone is quite anabolic.

Do not expect Masteron to provide much mass in kilograms. It is far away in this regard from testosterone or Dianabol, not to mention Anadrol, and it cannot provide the same strength gains that testosterone or trenbolone provides. But it does provide a clean and distinct anabolic impact, and it is also favorable when combined with almost any steroid, as we explain in its full review.

Superdrol is the methylated version of it.

A methyl group is added to the steroid in order to turn it orally active. The methyl group prevents the liver from deactivating it too quickly. The addition of the methyl group, as proven with many anabolic steroids, also adds to the potency and aggressive nature of the hormone.

As an example, Dianabol, which is one of the most aggressive and fast-acting steroids, especially milligram per milligram, is the methylated version of boldenone, which is a much milder — yet still effective — anabolic agent.

So, to some degree, Superdrol is “Masteron on steroids.” 🙂

The DEA Data: Why Superdrol Became a Legend

Superdrol, or methasterone, was synthesized in 1956, and its anabolic activity was reported in 1959. The most telling authoritative literature on Superdrol actually comes from the DEA, which refers to pharmacology data describing it as a potent anabolic agent with relatively weak androgenic activity.

The same document, relying on in-vivo models — real-life assays done on mammals — includes one report showing that Superdrol demonstrated four times the anabolic activity and one-fifth the androgenic activity of methyltestosterone when given orally.

The DEA also summarized data showing androgen-receptor affinity comparable to testosterone, along with greater activity in a transactivation assay.

To remind you, every steroid works through a receptor. The receptor binds to the steroid and introduces its signal into the cell. There, it commands the synthesis of proteins through what is referred to as transactivation.

So Superdrol binds to the androgen receptor as avidly as testosterone, yet it stimulates anabolic activity in the cell at a higher rate — a testimony to its very high anabolic potency.

And here is one of the most interesting parts: despite all of this potency, despite its chemistry, despite its activity, and despite its legendary reputation in bodybuilding, Superdrol has never been approved for medical use.

This is part of what makes its story so unusual. Some steroids came from clear medical backgrounds and later moved into bodybuilding culture. Superdrol took a much stranger path: it was born in the literature, disappeared into obscurity, then exploded during the prohormone era as something many people did not fully understand at the time.

Estrogen, Progesterone, and Cortisol: How Superdrol Creates Rock-Hard Muscles

The comprehensive DEA review also found that methasterone lacked the structural features needed for significant estrogenic, progestational, or corticosteroid activity. It then confirmed low binding affinity and low to no transactivation at the estrogen, progesterone, and glucocorticoid receptors.

Let’s go over these one by one.

Estrogenic activity — Superdrol is the methylated version of Masteron. Both are close relatives of DHT, which, unlike testosterone, cannot be converted into estrogen, the primary female hormone.

So, it is no surprise that Superdrol has no estrogenic effects. It simply cannot convert to estrogen, and by itself it does not activate the estrogen receptor.

Now for the interesting part: the DEA also noted a mouse assay reporting anti-estrogenic activity. So Superdrol does not just lack estrogenic effects; it actually counteracted estrogenic activity in that model.

This is worth noting because Masteron itself was used clinically in breast cancer treatment due to its anti-estrogenic effects. Due to these anti-estrogenic attributes, Masteron has a long-standing reputation as a hardening agent in bodybuilding and contest preparation. Its anti-estrogenic nature helps explain why it is so appreciated for producing a harder, drier, more sculpted physique.

Progesterone activity — Some steroids, particularly from the 19-nor family, such as Deca Durabolin and trenbolone, can carry some progestogenic activity. Progesterone is another female sex hormone, and when a steroid carries progestogenic activity, it can contribute to a softer look, water retention, libido issues, and estrogen-like side effects in sensitive users.

Masteron is free from these effects, and Superdrol appears to follow the same logic. This further explains why Superdrol is associated with a dry, shredded look rather than a watery or bloated appearance, which many steroids such as Dianabol, testosterone, or Deca are associated with.

Cortisol activity — Cortisol is known as the catabolic hormone. Many steroids achieve part of their anabolic effect by counteracting cortisol, and an agent such as Anadrol is especially associated with this phenomenon.

Superdrol lacks this effect. Its anabolic effect appears to come mainly from a very powerful direct impact through the androgen receptor, together with strong transactivation activity — meaning it helps trigger anabolic processes inside the muscle cell itself.

This is an important note.

Combining a steroid with such a direct anabolic mechanism with compounds that work strongly through other pathways, such as testosterone or trenbolone, may help explain why Superdrol has such a reputation for dramatic strength and recomposition effects.

Notice that we did not mention Anadrol here as a combination idea. Both Superdrol and Anadrol are harsh oral steroids with serious potential for liver stress, so combining them would generally be a poor idea, especially for any measurable length of time.

The Liver Question: Why Superdrol Must Be Respected

Now let’s talk about the part everyone knows is coming: the liver.

Superdrol is a 17-alpha methylated steroid, and this is exactly what makes it orally active. The methyl group helps the hormone survive the first pass through the liver, instead of being destroyed too quickly.

That is the magic.

But that is also the price.

The same feature that makes Superdrol powerful and orally active also makes it stressful on the liver. This does not mean every discussion about Superdrol needs to sound like a horror movie. Bodybuilding is full of compounds that require respect, knowledge, and proper judgment. But Superdrol is not one of those mild, forgiving agents that people can treat casually.

This is the real paradox of Superdrol.

The liver modification makes it stronger, faster, more aggressive, and more efficient — but it also puts it in the category of steroids that need serious caution. That is why experienced users speak about it with both admiration and respect. It can deliver a dramatic look, impressive strength, and a unique dry effect, but it is not a compound for reckless use or long, careless experiments.

This is also why the injectable version of Superdrol became such an interesting idea.
